Parmi ses rôles les plus célèbres au cours de sa longue carrière, on peut citer celui de Gerald O'Hara, le père de Scarlett O'Hara dans Autant en emporte le vent, celui du Doc Boone ivrogne dans La Chevauchée fantastique de John Ford, et celui de l'oncle Billy dans La Vie est belle. Mitchell fut la première personne à remporter un Oscar, un Emmy Award et un Tony Award.
